Abstract
To take full advantage of the exciting developments in confocal and multi-photon microscopy, it is important to have some understanding of light microscopy. It is not only useful in selecting the correct objective lens and microscope for the task at hand, but this knowledge will also allow one to interpret with greater confidence the images obtained. This chapter gives a brief introduction to light microscopy, with a strong emphasis on the practical aspects that you need to know about to take full advantage of the light microscope when working with a confocal or multi-photon microscope. The specific instrumentation required for confocal microscopy is dealt with in detail in the following chapter (Chapter 3 “Confocal Microscopy Hardware”, page 65).
