Creation of barley genotypes with complex resistance to edaphic stresses by methods of cell culture

Abstract

Barley genotypes resistant at the same time to acidity, aluminum toxicity, and osmotic stress were created by methods of cell culture. We developed optimal selective media for callus cultures containing 20–30 mg/L of Al3+ and 10% of polyethylene glycol (PEG) added in direct and reverse sequences. We observed an increase in the survival rate and the regenerative activity of callus after the reintroduction of genotype into the culture in vitro. We found six regenerant lines with crop yield significantly (by 10–43.2%) exceeding that of Bios 1 standard and original varieties, which was provided by higher productive tillering (28% higher) and kernel percentage (10% higher than in standard). The advantage of regenerant lines was especially obvious in high acidity provocative background (pH = 4.2) under abnormally hot and dry conditions of 2010.
