Genetic polymorphism of C4 in man and localisation of a structural C4 locus to the HLA gene complex of chromosome 6

Abstract

NATIVE C4, the fourth component of human complement (C4)1 has a molecular weight of 204,000 and is composed of three distinct polypeptide chains linked by disulphide bonds and non-covalent forces2. C4 migrates as a β1 globulin when subjected to electrophoresis in Agarose gel. Genetically determined structural polymorphism has been detected in the complement components C3 (refs 3 and 4), C6 (ref. 5) and properdin factor B (ref. 6). As well as their use in forensic science and population genetics, such polymorphisms are valuable tools for work on genetic linkage and chromosome mapping, as shown by the assignment of the factor B locus to chromosome 6, closely linked to HLA-B7,8. We report here similar findings with human C4.
