Effect of Physiological Age and Growth Regulators on Micropropagation of Aloe vera Followed by Genetic Stability Assessment

Abstract

Effect of physiological age and growth regulators on micropropagation of Aloe vera was investigated to develop an efficient protocol for in vitro propagation using shoot bud as explant. Maximum per cent establishment of 72.22 % was observed on MS medium supplemented with 10 µM BA, 5.0 µM Kn and 1.50 µM NAA. The best medium for the multiplication of microshoots was MS medium supplemented with 10 µM BA and 7.5 µM Kn followed by subculturing on same medium for six times at an interval of 4 weeks. Increase in number of microshoots per explant was observed with the increase in subculturing passages. After sixth subculturing 22.70 number of microshoots were obtained from the single explant which was four times more as compared to the first subculturing. The percent root induction was 77.78 % in the sixth subculturing as compared to 36.11 % in the first subculturing on MS medium supplemented with 0.04 % activated charcoal. Genetic stability of the in vitro grown plants was analysed by RAPD which indicated negligible variation.
