Abstract
We used 454 shotgun sequencing to locate microsatellite loci for use in molecular studies of the Florida mouse (Podomys floridanus). A total of 9,350 microsatellites were detected, from which a total of 23 were selected for screening and characterization. Of the initial 23 markers, 14 were retained based on amplification success and ease of scoring and further screened on 49 Florida mice sampled at four adjacent management units. The number of alleles per locus ranged from 4 to 12, with mean observed and expected heterozygosity values of 0.605 and 0.741, respectively. Three loci showed evidence of null alleles at both the management unit level and across combined samples, while two additional loci deviated from HWE when samples were pooled. No loci exhibited significant linkage disequilibrium.
