Abstract
A simple multiplex PCR method was developed for detection of the alleles H1 and Gro1-4 conferring resistance of potato to Globodera rostochiensis. SCAR markers TG689 and Gro1-4 linked to H1 and Gro1-4 genes respectively were amplified in the one PCR reaction. This allows to identify clones with resistance to G. rostochiensis originating from a single source or both of them. Effectiveness of this procedure for marker-assisted selection (MAS) was confirmed by 60 potato cultivars tested.
Resumen
Se desarrolló un método simple multiplex de PCR para la detección de los alelos H1 y Gro1-4 que confieren resistencia de la papa a Globodera rostochiensis. Se amplificaron los marcadores SCAR TG689 y Gro1-4 ligados a los genes H1 y Gro1-4 respectivamente en una reacción de PCR. Esto permite la identificación de clones con resistencia a G. rostochiensis originaria de una o ambas fuentes. Se confirmó la efectividad de este procedimiento para selección asistida con marcadores (MAS) probándolo en 60 variedades de papa.
