Resistance of Potato to Stem Infection by Phytophthora infestans and a Comparison to Detached Leaflet and Field Resistance Assessments

Abstract

To study the potato stem resistance to late blight, a new laboratory method has been established. Our aim was to understand relationships among resistance to P. infestans in stems and leaflets tested in laboratory and field resistance of plants. Sensitivity to P. infestans of stems and leaflets was investigated in 2004 and 2005 for fourteen cultivars, and in 2005 for fifteen breeding lines of various origins. We have performed a field assessment in 2005 for breeding lines and chosen cultivars. Laboratory tests were carried out twice a year, and their results correlated significantly. The correlation coefficients between leaflet and stem resistance data were significant and high (r ranged from 0.81 to 0.87). The correlation coefficient between Relative Area under the Disease Progress Curve (rAUDPC) values and stem resistance (r=−0.72) was higher than that between rAUDPC and leaflet resistance (r=−0.57). Laboratory assays for stem and detached leaflets resistance may be used for primary selection of breeding lines before their evaluation in field trials.

Resumen

Se ha establecido un nuevo método de laboratorio para estudiar la resistencia del tallo al tizón tardío. Nuestra meta fue entender las relaciones entre resistencia a P. infestans en tallos y folíolos probados en laboratorio y en resistencia de campo de plantas. La sensibilidad a P. infestans de tallos y folíolos se investigó en 2004 y 2005 para catorce variedades, y en 2005 para quince líneas mejoradas de varios orígenes. Desarrollamos un análisis de campo en 2005 para líneas de mejoramiento y variedades selectas. Las pruebas de laboratorio se llevaron a cabo dos veces al año, y sus resultados se correlacionaron significativamente. Los coeficientes de correlación entre los datos de resistencia de folíolos y tallo fueron significativos y altos (r fluctuó de 0.81 a 0.87). El coeficiente de correlación entre los valores del Área Relativa bajo la Curva de Progreso de la Enfermedad (rAUDPC) y la resistencia de tallo (r=−0.72) fue más alta que la de entre rAUDPC y la resistencia del folíolo (r=−0.57). Los ensayos de laboratorio de resistencia de tallo y de folíolos separados pudiera usarse para una selección primaria de líneas de mejoramiento antes de su evaluación en ensayos de campo.
