Establishment and Validation of a Time-Resolved Fluoroimmunoassay for Chlorpromazine

Abstract

The objective of this study is to establish a highly sensitive, time-resolved fluoroimmunoassay (TRFIA) for the rapid detection of chlorpromazine (CPZ). The antigen CPZ-OVA was used to coat a microtiter plate to compete with free chlorpromazine for binding to the anti-chlorpromazine monoclonal antibody. Europium (Eu3+)-labeled goat anti-mouse antibody was used as the tracer. The labeling rate of Eu3+-labeled goat anti-mouse antibody was 10.5 Eu3+/antibody. The average intra- and inter-batch coefficients of variation were 4.0% and 6.4%, respectively, and the average recovery rate was 90.27%. The sensitivity was 0.2 μg/L over a measurable range of 0.2–50 μg/L. The ED20, ED50, and ED80 were 1.17, 4.85, and 17.12 μg/L, respectively. CPZ-TRFIA is one of the most sensitive methods for CPZ detection. This analytical method demonstrates good stability and a wide measurable range, demonstrating its potential for practical applications.
