Comparison of nuclease activities of binuclear metal complexes with N6O coordinating heptadentate ligands: [M2L(OAc)2]+ (M = Ni, Mn, L = 2,6-bis(((2-(dimethylamino)ethyl)(pyridin-2-ylmethyl)amino) methyl)-4-methoxyphenol)

Abstract

Four complexes, [Ni2L(OAc)2](PF6) (1), [Ni2L(OAc)2](BPh4) (2), [Mn2L(OAc)2](PF6) (3) and [Mn2L(OAc)2](BPh4) (4) (L = N,N,N′,N′-2,6-bis(((2-(dimethylamino)ethyl)(pyridin-2-ylmethyl)amino)methyl)-4-methoxyphenol), have been synthesized. Single crystal diffraction reveals that the metal atoms in all four complexes have a distorted octahedral geometry. The interactions of the complexes with calf thymus DNA (CT-DNA) have been investigated by UV absorption, fluorescence spectroscopy, circular dichroism spectroscopy and viscosity measurements, and the most likely mode of CT-DNA binding for these complexes is identified as moderate interaction. Furthermore, the complexes are capable of promoting DNA cleavage through an oxidative pathway, involving hydroxyl radical, singlet oxygen or singlet oxygen-like species.
