Karzinogenese und hereditäre Kolonkarzinome

Zum Thema

Belegt durch eine große Anzahl experimenteller Daten erlangte die Theorie der somatischen Mutation als wesentliches Ereignis bei der Entstehung von Tumoren im letzten Jahrzehnt eine Schlüsselrolle. Mutationen können zur Aktivierung von Onkogenen und Inaktivierung der Funktion von Tumorsuppressorgenen führen [5]. Die Analyse dieser tumorspezifischen genetischen Veränderungen erbrachte fundamentale Erkenntnisse nicht nur über die Tumorentwicklung, sondern auch über die normale Zellproliferation. Die Ätiologie der zugrunde liegenden Mutationen bleibt jedoch weiterhin unklar. Bis heute ist nicht entschieden, ob genotoxische Agenzien oder einfache spontane Fehler während der DNA-Replikation einen größeren Einfluss auf die Genese dieser Mutationen haben.
