Abstract
Chronic diabetes alters sympathetic modulation of the cardiovascular system. In the present work, we examined if the cardiovascular system also demonstrates an impaired responsiveness to sympathetic control. The effects of streptozotocin-induced diabetes on pressure responses to noradrenaline or isoproterenol infusion of diabetic ratsin vivo are studied. Systolic and diastolic pressures were recorded through a cannula implanted in the right carotid artery. Increasing doses of noradrenaline or isoproterenol were infused through a catheter implanted in the left jugular vein. The dose-response curves for the effects of noradrenaline on blood pressure were altered in streptozotozin-induced diabetic rats. Noradrenaline induced a statistically significant higher increase of both systolic and diastolic pressure in control than in diabetic rats. The maximum depressor response of systolic pressure to isoproterenol was lower in diabetic than in control animals. Diabetes fully abolishes the effect of β-adrenoceptor stimulation on diastolic blood pressure. The present results demonstrate that streptozotocin-induced diabetes reduces systolic and diastolic arterial pressure, and diminishes the arterial pressure reactivity to sympathetic stimulation.
Resumen
La diabetes crónica altera la influencia del sistema nervioso simpático sobre el control circulatorio. En este trabajo, se estudia si el sistema cardiovascular tiene también alterada su capacidad de respuesta a la estimulación simpática. Para ello, se estudianin vivo los efectos de la infusión de noradrenalina e isoproterenol sobre las respuestas cardiovasculares de ratas diabéticas por inyección de estreptozotocina. La presión arterial sistólica y diastólica se registran mediante una cánula implantada en la arteria carótida derecha. Dosis crecientes de noradrenalina e isoproterenol se administran por vía endovenosa mediante un catéter implantado en la vena yugular izquierda. Las curvas dosis-respuesta para los efectos de la noradrenalina sobre la presión arterial se ven alteradas por la diabetes. La noradrenalina produce un incremento de las presiones sistólica y diastólica significativamente mayor en animales control con respecto a los animales diabéticos. La máxima respuesta depresora de la presión sistólica al isoproterenol es menor en animales diabéticos que en sanos. La diabetes elimina completamente el efecto de la estimulación de los adrenoceptores β sobre la presión diastólica. En suma, la diabetes inducida por estreptozotocina reduce la presión arterial sistólica y diastólica, y disminuye la reactividad cardiovascular a la estimulación simpática.
