Abstract

Site-directed mutagenesis techniques have been applied to theglyA gene ofE. Coli coding for serine hydroxymethyltransferase, in order to substitute amino acid residues known to be at the active site of the enzyme. Two high efficiency mutagenesis methods have been used that specifically select,in vivo orin vitro, mutant DNA with respectto the wild-type one.

Sono state applicate tecniche di mutagenesi sito-specifica al geneglyA diE. coli codificante per la serina idrossimetiltrasferasi, al fine di sostituire alcuni residui aminoacidici che si suppone siano implicati nel meccanismo ďazicne delľenzima. Nel corso degli esperimenti sono stati utilizzati due metodi ad alta efficienza di mutagenesi, basati sulla specifica selezione,in vivo o in vitro, del DNA mutato rispetto a quello selvatico.
