Abstract
Agrobacterium tumefaciens-induced tumor formation in plant cells involves transfer of a defined segment of DNA (T-DNA) from the bacterium into plant cells. This process is known to involvevir genes, 25-bp-long T-DNA border sequences and additional sequences adjacent to border sequences called overdrive sequences. It is also known that thevir D gene-encoded endonuclease causes cleavage at the border sequences. In this report we show that a synthetic 25-bp border sequence is sufficient for cleavage and that overdrive sequences are not necessary for the cleavage catalysed byvirD endonuclease.
