Summary
After hydrocortisone application to the dorsal epidermis of mice, the mitotic rate was reduced more than 60% within 5 h. Thymidine-3H incorporation into epidermal DNA was also inhibited, with a maximum reduction at 12 h with all doses tested. A second decrease in the mitotic rate followed the inhibition of DNA synthesis. Thus, cells in G1 and in G2 are susceptible to the inhibitory effects of hydrocortisone.
The combined effects of hydrocortisone and epidermal chalone on DNA synthesis were not greater than the effect of either agent alone. When hydrocortisone was applied prior to treatment with croton oil, the extent of the croton oil-induced increase in DNA synthesis was reduced.
