Behavior of testicular and ovarian fragments of the newt pleurodele cultured in serum-free medium

Summary

A method has been developed for culturing testes and ovaries of the young salamanderPleurodeles waltlii Michah. in a serum-free medium for 3 wk. Testicular and ovarian fragments placed in “petriperm” dishes were cultured at pH 7.2 to 7.4 in bicarbonate buffered Medium 199 (M199), brought to osmolarity of 240 mOsm, supplemented with antibiotics. Incubation was carried out at 20° C in a humidiffed atmosphere of 95∶5% air and CO2. After 3 wk of culture in hormone-free medium, germ cells showed a normal degeneration in testicular explants, whereas the ovarian explants retained normal histological characteristics as well as transcription and translation activities. Addition of insulin (5 μg/ml) to this medium produced striking effects on the testicular explants, particularly on the spermatogonial mitoses. The addition of 1 U/ml of follicle stimulating hormone (FSH) resulted in maintenance of complete spermatogenesis. In contrast, insulin or FSH produced no effect on the ovarian explants.
