Abstract
Microbiological methods for the evaluation of essential e.g. vital compounds have been propagated for the first time since the beginning of this century. They reached practicability, however, not before the time-space between 1940 and 1950, and were then widely used for the determination of amino acids and vitamins.
An exact knowledge of the nutritive demand of the test organisms is prerequisite for their application in the quantitative analysis in order to exclude uncontrolled growth stimulations and thus adulterations of the results by components other than those which are to be determined. As test organisms—which especially should not be pathogenic for practical use—mainly serve lactic acid producing bacteria, yeasts and mutants of Neurospora.
It is stated that microbiological methods can be applied with profit in those cases where it is necessary to quantitates only a single or a few of essential components in a great number of samples, a problem often arising when the nutritional and physiological value has to be destined.
A short description of the ‘classical’ methods of microbiological assay is followed by the review of newer modifications which permit the partial- or full-automated performance of microbiological methods. This is shown for the analysis of essential amino acids as an example. Finally the possibility of estimating the biological value of proteins is shortly discussed.
Zusammenfassung
Mikrobiologische Verfahren zur Bestimmung von essentiellen, d.h. lebensnotwendigen Verbindungen reichen in ihren ersten Anfängen bis zur Jahrhundertwende zurück. Sie konnten jedoch erst in den Jahren zwischen 1940 and 1950 Praxisreife erreichen und wurden dann in großem Umfang zur Bestimmung von Aminosäuren und Vitaminen eingesetzt.
Voraussetzung für ihre Anwendung in der quantitativen Analytik war eine genaue Kenntnis der Nährstoffansprüche der verwendeten Testorganismen. um eine unkontrollierte Wachstumsstimulation und damit Verfälschungen der Ergebnisse durch andere als die zu bestimmende Komponente auszuschließen. Als Testorganismen—die für ihre praktische Verwendung insbesondere nicht pathogen sein dürfen—dienen hauptsächlich Milchsäurebakterien, Hefen und Neurospora-Mutanten.
Es wird festgestellt, daß mikrobiologische Verfahren immer dort Vorteil eingesetzt werden können, wo es darauf ankommt, in einem großen Probenmaterial eine einzige oder wenige essentielle Komponenten zu bestimmen, eine Problemstellung die bei der Ermittlung des ernährungsphysiologischen Wertes häufig auftritt.
Nach einer kurzen Darstellung der ‘klassischen’ Methodik mikrobiologischer Bestimmungen wird auf neuere Modifikationen eingegangen, die es gestatten, mikrobiologische Verfahren auch teil- bzw. vollautomatisiert durchzuführen. Dies wird an einem Beispiel aus der Aminosäuren-Analytik dargestellt. Abschließend wird auf die mikrobiologische Bestimmung der biologischen Wertigkeit von Eiweißkörpern kurz eingegangen.
