Abstract
A sensitive method for identifying and counting osteoclasts has been developed, using the intense succinic dehydrogenase activity demonstrable in these cells. The method was used to compare the time relation of the changes in blood calcium and osteoclast numbers in the mouse femur after the administration of 1 U PTE/mouse. An age-related decrease in osteoclast count was found. The earliest significant rise in osteoclast numbers, after the administration of PTE, occurred much earlier than has been previously reported. While the results at present indicated that the hypercalcaemic response to an intravenous injection of PTE was complete before an appreciable rise occurred in the osteoclast count, they did not exclude the possibility that the osteoclasts exert a hypercalcaemic effect by an increase in activity rather than in numbers. Alternatively, the results may indicate that osteoclasts are not involved in the mediation of the hypercalcaemic response to PTE.
Résumé
Une méthode précise pour identifier et compter les ostéoclastes été mise au point en utilisant leur grande activité en déshydrogénase succinique. Le rapport chronologique des changements du calcium sanguin et le nombre des ostéoclastes ont été comparés au niveau du fémur de souris après administration de 1 U PTE/souris. Une diminution en ostéoclastes est observée en fonction de l’âge. La première augmentation significative du nombre des ostéoclastes, après administration de PTE, a été observée bien plus précocément que dans les publications antérieures. Alors que les résultats actuels indiquent que la réponse hypercalcémique à une injection intraveineuse de PTE s’observe avant l’augmentation notable du nombre des ostéoclastes, la possibilité que les ostéoclastes exercent une action hypercalcémique par augmentation de leur activité plutôt que par leur nombre augmentée, n’est pas exclue. De même il se pourrait que les ostéoclastes interviennent dans la réponse hypercalcémique à la PTE.
Zusammenfassung
Es wurde eine empfindliche Methode zur Identifizierung und Zählung von Osteoclasten entwickelt, wobei die intensive Succino-Dehydrogenase-Aktivität, welche in diesen Zellen nachgewiesen werden kann, verwendet wurde. Mit dieser Methode konnten die Veränderungen im Blutcalcium und in den Osteoclastenzahlen im Mäusefemur nach der Verabreichung von 1 E Parathyreoidea-Extrakt (PTE) per Maus als Funktion der Zeit ausgewertet werden. Eine altersabhängige Abnahme der Osteoclastenmenge wurde festgestellt. Der früheste signifikante Anstieg der Osteoclastenzahlen nach der Verabreichung von PTE erfolgte viel früher als bis dahin angenommen. Die jetzigen Ergebnisse deuten darauf hin, daß die hypercalcämische Reaktion auf eine intravenöse Injektion von PTE abgeschlossen ist, bevor ein feststellbarer Anstieg der Osteoclastenzahl erfolgt; sie schließen jedoch die Möglichkeit nicht aus, daß die hypercalcämische Wirkung der Osteoclasten durch eine Erhöhung der Aktivität anstatt der Anzahl hervorgerufen werden kann. Andererseits können die Ergebnisse auch bedeuten, daß die Osteoclasten an der Übermittlung der hypercalcämischen Reaktion auf PTE nicht beteiligt sind.
