Abstract
Ultrastructural changes of myocardial and endothelial cells in the microvasculature of the rat heart following global ischemia were observed by transmission electron microscopy. The hearts of Wistar rats were made ischemic by clamping the aorta, for 0, 15, 60, and 120 min in groups I, II, III and IV, respectively. Mitochondria in myocardial cells appeared irregular in shape and were scattered in groups III and IV. Glycogen granules decreased, and myofibrils were loosened in groups III and IV. Occasional cytoplasmic protrusions into the lumen were seen in the cytoplasm of endothelial cells from group II. In group III, protrusions were frequently observed and vacuoles appeared in the cytoplasm of the endothlial cells. In group IV, collapsed cytoplasm of the endothelial cells appeared in the lumen, and vacuoles in the cytoplasm were enlarged. Pinocytotic vesicles of the endothelial cells decreased in number in groups III and IV. From this ultrastructural evidence, it is concluded that myocardial cell injury may lag behind microvascular damage during global ischemia of the rat heart.
