Summary
The main sources of error in chromatographic analysis are described, particular attention being drawn to variable bias. This is one of the major potential sources as it is not easily detected. A comparison is made between the charateristics of the three basic methods of calculating results, normalization, constant volume injection, and internal standard. Despite its popularity normalization has many disadvantages and the presentation of results as ratios of compounds to one another is suggested in preference to normalization.
Experimental evidence is given to demonstrate that no commercial chromatograph tested gives relative response factors within acceptable limits of published data and that no commercial flame ionisation detectors are truly linear over much of their range. Calibration of individual chromatographs with synthetic mixtures covering the range of concentrations expected is advocated as being the best way to obtain accurate results. If individual calibration is used, the use of peak area as the measure of peak size is not essential, and experimental evidence is given to show that better repeatability is obtained from peak height measurement than from peak area measurement.

Es werden die wesentlichsten Fehlerquellen der chromatographischen Analyse beschrieben, wobei variablen systematischen Fehlern besondere Beachtung geschenkt wurde. Da diese nicht leicht erkannt werden können, stellen sie möglicherweise wichtige Fehlerquellen dar. Zwischen den Eigenschaften der drei Grundmethoden zur Berechnung der Ergebnisse wird ein Vergleich angestellt: Normierung, Dosierung konstanten Volumens und innerer Standard. Obwohl sie allgemein eingeführt ist, hat die Normierung viele Nachteile. Es ist besser, die Ergebnisse relativ bezogen auf eine Substanz anzugeben, als allgemein auf 100% zu normieren.
Es wird experimentell bewiesen, daß keiner der getesteten handelsüblichen Chromatographen relative Response-Faktoren innerhalb annehmbarer Grenzen von veröffentlichten Daten liefert, und daß keiner der (getesteten) handelsüblichen Flammenionisationsdetektoren wirklich linear über seinen angegebenen Bereich ist. Die Eichung der einzelnen Chromatographen mit synthetischen Mischungen (die den erwarteten Konzentrationsbereich überstreichen) ist der beste Weg, um genaue Ergebnisse zu erhalten. Wenn diese individuelle Eichung benutzt wird, ist die Verwendung der Peakfläche als Meßgröße nicht wesentlich. Experimentell wird gezeigt, daß durch Messen der Peakhöhe eine bessere Wiederholbarkeit erreicht wird als durch Ermitteln der Peakfläche.

Ce texte décrit les principales sources d'erreurs en analyse chromatographique et, en particulier, attire l'attention sur les erreurs systématiques fluctuantes. Leur identification n'étant pas aisée, ce sont elles qui constituent probablement les sources d'erreur les plus importantes. On compare les caractéristiques des trois méthodes fondamentales permettant de calculer le résultat: normalisation, injection d'échantillons de volumes constants, et étalon interne. Bien qu'elle soit généralement adoptée, la normalisation présente beaucoup d'inconvénients, et on suggère de donner les résultats sous forme de concentrations relatives plutôt que sous forme normalisée.
Les expériences décrites montrent qu'aucun des chromatographes commerciaux étudiés ne donne de facteurs de réponse relatifs qui soient en accord acceptable avec les valeurs publiées dans la littérature. De même, aucun détecteur à ionisation par flamme commercial n'est vraiment linéaire dans la plus grande partie de son domaine de réponse. L'étalonnage effectué pour chaque chromatographe à l'aide de mélanges synthétiques couvrant tout le domaine de concentrations attendu est le meilleur moyen d'obtenir des résultats précis.
