Cells ofSchizosaccharomyces pombe from the stationary growth phase show only a low capacity for the uptake of xanthine. In the presence of 2 μM xanthine, the initial uptake rate is found in the range of 6 · 10−21 Moles × Cells−1 × Min−1 at 30 °C.

Preincubation in glucose solution results in a pronounced stimulation of the uptake rate.

The stimulating effect of the glucose pretreatment is repressed by cycloheximide, ammonium ions and amino acids.

The substrate dependency of the uptake rate shows a saturation kinetics. The Km-value has been determined to 6 μM. The temperature optimum is found in the range of 30 °C.

Xanthine is only slightly accumulated within the cells against the external concentration. Most of the external xanthine label is stored up in form of xanthosine and guanosine as well as incorporated in nucleic acids.

Zellen vonSchizosaccharomyces pombe aus der stationären Wachstumsphase zeigen nur eine begrenzte Aufnahmefähigkeit für Xanthin. In Gegenwart von 2 μM Xanthin beträgt die initiale Aufnahmegeschwindigkeit bei 30 °C 6 · 10−21 Mol × Zelle−1 × Min−1.

Eine Präinkubation dieser Zellen in Glucoselösung bewirkt eine starke Stimulation der Aufnahmegeschwindigkeit.

Der stimulierende Effekt der Glucosevorbehandlung wird durch Cycloheximid, Ammoniumionen und Aminosäuren unterdrückt.

Die Kinetik des Transportsystems weist eine Substratsättigung auf. Der Km-Wert beträgt 6 μM. Das Temperaturoptimum liegt bei 30 °C.

Xanthin wird in der Zelle nur geringfügig gegenüber der Außenkonzentration akkumuliert. Die Radioaktivität des extern angebotenen Xanthins wird hauptsächlich in Xanthosin und Guanosin gespeichert, zum Teil auch in Nukleinsäuren eingebaut.
