The influence of the calcium-complexes of the aminopolycarboxylic acids (APA). — HEDTA, EDTA, CDTA, DTPA and TTHA — on the excretion and the distribution of zinc after the application of65ZnCl2 (with carrier) and of the chelating agents mentioned was followed in acute experiments in the rats.
These chelating agents applied simultaneously with ZnCl2 increase the excretion of zinc into the urine and decrease the excretion of zinc into the feces. The total amount of zinc excreted from the organism is increased. CaDTPA, CaHEDTA and CaTTHA appear the most efficient. The effect of APA decreases with the lengthening of the time interval between application of Zn and the chelating agents. CaDTPA and CaTTHA are the most effective agents again.
In the urine of the rats given ZnCl2 (non-radioactive) and Ca-complexes of APA, Ca- and Zn-complexes of APA as well as the ion-associates of these complexes with potassium and magnesium may be proven by paper chromatography.
Zusammenfassung
An Ratten wurde experimentell der Einfluß der Calciumkomplexe der Aminopolycarbonsäuren (APS) — HÄDTA, ÄDTA, CDTA, DTPA und TTHA — auf die Ausscheidung des Zinks und dessen Verteilung im Organismus nach einmaliger Applikation von65ZnCl2 (mit Träger) und der oben erwähnten Chelatbildner studiert. Alle genannten Chelatbildner erhöhten bei gleichzeitiger Verabreichung von ZnCl2 die Ausscheidung des Zinks im Urin und setzen diese im Stuhl herab. Die gesamte Menge des ausgeschiedenen Zinks aus dem Organismus ist erhöht. Am wirksamsten erweisen sich CaDTPA, CaHÄDTA und CaTTHA. Wird das Zeitintervall zwischen der Applikation von ZnCl2 und der Chelatbildner verlängert, so nimmt die Wirkung dieser Stoffe ab. CaDTPA und CaTTHA sind abermals die wirksamsten. Die chromatographische Analyse des Harns von Ratten, denen ZnCl2 (nicht aktiv) und die Ca-Komplexe der APS appliziert wurden, zeigt, daß im Urin die Ca- und Zn-Chelate der APS und Ionenassoziate dieser Komplexe mit Kalium und Magnesium vorhanden sind.
