Abstract
A study was made of the glycosaminoglycans of dentine and predentine in rachitic puppies. These two tissues were obtained by a microdissection procedure, digested with papain and the glycosaminoglycans precipitated with cetylpyridinium chloride. The glycosaminoglycans were analysed by infrared spectrophotometry and by chromatography on cetylpyridinium chloride, Ecteola and Dowex 50. The results indicate that the major polysaccharides in dentine and predentine are galactosaminoglycans tentatively identified as chondroitin sulphates. The concentration of polysaccharide—expressed as hexosamine—was 0.06% organic dry weight in dentine; the corresponding figure for predentine was 0.12%. The results, which indicate, furthermore, that there is relatively more chondroitin-6-sulphate in predentine than in dentine, are discussed briefly in relation to mineralization.
Résumé
Une étude des glycosaminoglycanes de la dentine et prédentine est réalisée chez de jeunes chiens rachitiques. Ces deux tissus, obtenus après microdissection, sont digérés à la papaïne et les glycosaminoglycanes sont précipités par le chlorure de cetylpyridinium. Les glycosaminoglycanes sont analysés par spectrophotométrie infra-rouge et par chromatographie sur chlorure de cetylpyridinium, Ecteola et Dowex 50. Les polysaccharides principaux de la dentine et de la prédentine sont les galactosaminoglycanes, probablement sous forme de sulfates de chondroitine. La concentration du polysaccharide—exprimé en hexosamine—est de 0.06% de poids sec organique dans la dentine et de 0.12% pour la prédentine. Le fait qu'il y ait relativement plus de chondroitine-6-sulfate dans la prédentine que dans la dentine est discuté sous l'angle de la minéralisation.
Zusammenfassung
Die Glykosaminoglycane von Dentin und Prädentin wurden bei rachitischen jungen Hunden untersucht. Diese zwei Gewebe wurden durch Mikrodissektionsverfahren erhalten, dann mit Papain verdaut und die Glykosaminoglycane mit Cetylpyridinchlorid gefällt. Diese wurden durch Infrarotspektrophotometrie und durch Chromatographie auf Cetylpyridinchlorid, Ecteola und Dowex 50 analysiert. Die Resultate weisen darauf hin, daß die wichtigsten Polysaccharide in Dentin und Prädentin Galactosaminoglycane sind, welche vorläufig als Chondroitinsulfate identifiziert wurden. Die Polysaccharidkonzentration — als Hexosamin ausgedrückt — betrug bei Dentin 0,06% des organischen Trockengewichts; bei Prädentin betrug sie 0,12%. Die Ergebnisse, welche weiterhin andeuten, daß im Praedentin verhältnismäßig mehr Chondroitin-6-Sulfat als Dentin vorhanden ist, werden kurz in ihrer Beziehung zur Mineralisation diskutiert.
