Synthesis and pharmacological properties of [1-N4-dimethyl-asparagine]-angiotensin II

Summary

[1-N4-Dimethyl-asparagine]-angiotensin II was synthesized by Merrifield's solid-phase procedure. The analogue gave rat blood pressure about 70% relative potency to Hypertensin (Ciba). Rabbit aorta strips gave intrinsic activity αE=1, a PD2 of 6.92±0.09 and an affinity relative to [Asn1]-angiotensin II of 6.5%.

  6. Boc-amino acids were purchased from Protein Research Foundation (Japan) and are of the L-configuration. The following side chain protected Boc-amino acids were used: Nα-Boc-(Nim-tosyl)-histidine, Boc-(O-oBr-Z)-tyrosine, Nα-Boc-(Nω-nitro)-arginine and Nα-Boc-(N4-dimethyl)-asparagine, which was prepared from α-benzyl L-aspartate in a similar manner to that described for N2-carbobenzoxy-(N5-dimethyl-L-glutamine (Th. Caplaneris, Tetrahedron34, 969 (1978)).
