Following the i.m. injection of 400 mgα-methyldopa/kg body weight there appears a fluorescence inside the fibres of the muscles of the abdominal wall and the diaphragm of the rat. This fluorescence is specific for catecholamines and related compounds. It appears substantially more abundant inside the thin than the thick fibres.

In vitro-experiments with the diaphragm (300 mgα-methyldopa/l Krebs-Henseleit solution, 18°C) reveal the same picture which can be accentuated by electrical stimulation, especially within the thin fibres.

The recordings of the slow and fast component of the isometric contraction show the favoured reduction of the slow component as a consequence of the application ofα-methyldopa. This effect is the same during stimulation of the nerve or that of the muscle after blockade of the neuro-muscular transmission.

The results lead to the conclusion thatα-methyldopa exerts a direct action on the skeletal muscular system and predominately its thin fibres. It should be taken into account that the efficiency ofα-methyldopa under the treatment of hyperkinetic states would not only be the result of a central action of this substance but of a peripheral one too.
Zusammenfassung
In den Muskelfasern der Bauchmuskulatur und des Zwerchfells der Ratte tritt nach i. m. Gabe von 400 mg L-α-Methyldopa/kg eine katechol-aminspezifische Fluorescenz auf, die von einer unspezifischen Eigenfluorescenz der Muskulatur gut abzugrenzen ist. — Die spezifische Fluorescenz ist in den dünnen Muskelfasern wesentlich stärker als in den dicken.

Bei Untersuchung des Zwerchfells in vitro (300 mg L-α-Methyldopa/l Krebs-Henseleit-Lösung, 18°C) ist nach 1 Std ein gleichartiges Fluorescenzbild festzustellen. Zusätzliche elektrische Reizung verstärkt die Fluorescenz in den dünnen Fasern. Einstündige Inkubation in L-α-Methyldopa-haltigem Medium intensiviert die Phosphorylasereaktion in den dünnen Fasern. Bei gleichzeitiger Reizung wird der Phosphorylasenachweis wieder schwächer, während die PAS-Reaktion zum Glykogennachweis kräftiger ausfällt.

Bei Registrierung der schnellen und langsamen Komponente der isometrischen Kontraktion des Rattenzwerchfells fällt nach der Anwendung von L-α-Methyldopa vor allem die Reduzierung der langsamen Komponente auf. Diese Wirkung tritt sowohl bei indirekter als auch bei direkter Reizung nach Blockierung der neuromuskulären Transmission ein.

Die Befunde lassen darauf schließen, daß L-α-Methyldopa eine direkte Wirkung auf die Skeletmuskulatur und insbesondere auf deren dünne Fasern ausübt. Die Beurteilung der Wirkung vonα-Methyldopa bei Hyperkinesen sollte daher nicht nur einen zentralen, sondern auch einen peripheren Wirkungsmechanismus in Betracht ziehen.
