Recovery of lymphocytes and epithelioid cells from fluids used to flush the uteri and oviducts of superovulated cattle and preparation of monolayer cultures from epithelial cells of oviduct and uterus

Summary

A Ficoll-gradient method to recover separately lymphocytes and sedimented cells from uterotubal flush fluid of cattle is described. Lymphocytes could be cocultured for virus isolation or used for histopathological studies, or both. Epithelioid cells, most likely of oviductal origin, were grown in monolayer cultures to initiate cell lines and isolate viruses. This technique could be applied to other situations that require the growth of bovine oviduct epithelial cells in vitro.
