Comparative pharmacokinetics of sisomicin after intravenous injection, intravenous infusion and intramuscular injection in man

Summary

The parmacokinetics of sisomicin were determined in ten subjects after intravenous injection, intravenous infusion over a 35 to 45 minute period and after intramuscular injection of a single dose of 1 mg/kg respectively. The plasma elimination curve of sisomicin after i.v. administration revealed a bi-exponential decrease. The α-phase T/2 averaged approximately ten minutes after intravenous injection of sisomicin. The postdistributive plasma elimination halflives were 129 min after i. v. injection, 145 min after i. v. infusion and 161 min after i. m. injection. The apparent distribution volume after i. v. injection averaged 147 ml/kg. Urinary recovery rates were 57–80% after four hours and 75–100% after 24 hours.

Zusammenfassung

Das pharmakokinetische Verhalten von Sisomicin wurde bei je zehn Versuchspersonen nach intravenöser Injektion und Infusion und nach intramuskulärer Injektion einer Einzeldose von 1 mg/kg untersucht. Die Plasmaeliminationskurve von Sisomicin zeigte einen bi-exponentiellen Verlauf. Für die α-Phase wurde nach intravenöser Injektion eine Sisomicin-Halbwertzeit von rund zehn Min. ermittelt. Für die postdistributive β-Phase wurden durchschnittliche Halbwertzeiten von 129 Min. nach intravenöser Injektion, 145 Min. nach intravenöser Infusion und von 161 Min. nach intramuskulärer Verabreichung errechnet. Das virtuelle Verteilungsvolumen war im Durchschnitt 147 ml/kg. Nach vier Stunden waren bereits 57–80 %, nach 24 Stunden 75–100 % der injizierten Dosis im Urin nachweisbar.
