Summary
Extensive recent research has established that Prostaglandins (PG) are a widespread new class of biomediators. From polyunsaturated fatty acids PGs are formed enzymatically upon a variety of stimuli and then act at low concentrations. Participation has been shown so far for inflammation and pain, gastric secretion, various regulatory functions and others.
Total synthesis has been achieved on a number of routes. Structure variations aim at a greater specificity and a less rapid metabolistic breakdown. Therapeutic possibilities are indicated.
Zusammenfassung
In intensiver anhaltender Forschung wurden Prostaglandine (PG) als eine neue Klasse von Biomediatoren erkannt. Sie entstehen aus polyungesättigten Fettsäuren, wobei das Enzymsystem durch eine Vielfalt von Reizen angeregt wird. In niedriger Konzentration sind PG bei verschiedenartigen physiologischen Prozessen beteiligt: Entzündung, Schmerz, Sekretion des Magensaftes und einer Reihe von Regulationsfunktionen. Mehrere Totalsynthesen erlauben einen guten Zugang zu den natürlichen PG und einer großen Zahl von Strukturvariationen, die mit dem Ziel größerer Spezifität und weniger raschen metabolistischen Abbaus hergestellt wurden. Therapeutische Einsatzmöglichkeiten zeichnen sich ab.
