Intestinal disaccharidases in rats treated with 5-fluoro-uracil

Summary

Activities of rat intestinal disaccharidases are decreased following treatment even with low doses of 5-fluorouracil. Higher doses of the cytostatic drug cause morphological alterations of intestinal mucosa accompanied by a marked drop in enzyme activities. After a single dose of 5-fluoro-uracil, reduced disaccharidase activities are maintained for a couple of days even after morphological restitution of the mucosa.

Malabsorption of disaccharides associates with cytostatic treatment should be paid more attention also in man.

Zusammenfassung

5-Fluorouracil führt schon in relativ niedriger Dosierung zu einer Verminderung der Disaccharidase-Aktivitäten in der Dünndarmschleimhaut der Ratte. Bei höheren Dosierungen kommt es neben morphologischen Veränderungen der Darmmucosa zu einem erheblichen Abfall der Disaccharidase-Aktivitäten, die nach einer Einzeldosis auch nach morphologischer Restitution noch über Tage vermindert sein können. Der therapeutisch induzierten Disaccharid-Malabsorption sollte auch bei der zytostatischen Behandlung des Menschen größere Aufmerksamkeit geschenkt werden.
