Cyropreservation of parathyroid tissue: Examination of a simplified method

Summary

A simplified, less expensive cryopreservation technique for parathyroid tissue was examined in animal study and on human tissue in vitro. More than 3/4 of all replantations in the rat were succesful after cryopreservation. After replantation of one single gland only 1/3 of all animals attained normal parathyroid function. This fact shows the importance of the quantity of transplanted viable tissue. Viability of human parathyroid tissue after cryopreservation was 80–90%. The PTH-production of human parathyroid tissue in vitro after cryopreservation and the suppressibility in different calcium concentrations were similar to normal fresh tissue. All our results prove a good functioning of this simplified cryopreservation method.

Zusammenfassung

Eine vereinfachte und billigere Methode der Kryopreservation von Nebenschilddrüsengewebe wird im Tierversuch und an menschlichem Gewebe geprüft. Gber 3/4 aller Replantationen waren nach Kryopreservation bei Ratten erfolgreich. Wurde jeweils nur eine Drüse transplantiert, sank die Erfolgsquote auf 1/3, was darauf hinweist, daß die Menge des transplantierten Gewebes eine wesentliche Rolle spielt. Das kryopreservierte menschliche Nebenschilddrüsengewebe wies eine Vitalitätsrate von 80–90% auf. Die PTH-Produktion von kryopreserviertem Gewebe in vitro, sowie die Suppressibilität in verschiedenen Calciumkonzentrationen entsprechen dem Verhalten von frischem Gewebe. Die hier geprüfte Methode der Gefrierkonservierung kann anhand der Resultate als tauglich angesehen werden.
