Susceptibility of rabbit kidney cells of various passage levels to infection with SV-40 virus

Abstract

Rabbit kidney cells of increasing passage levels passed through various stages of response to SV-40 virus inoculation: during the first 13 to 24 passages there was a striking decrease in incubation time, i.e., the time until appearance of cytopathic effects, and lytic CPE always preceded transformation by several days. Then this pattern changed, lytic CPE appeared later, and finally did not appear at all, and transformation became the only indication of SV-40 infection.

Transformation was recognized by: 1. focal appearance of small epithelioid cells, 2. loss of contact inhibition, 3. enhanced growth rate, 4. higher acidity of culture medium, as well as 5. development of “T” antigen and almost complete absence of virion antigen as measured by complement fixation and staining with fluorescent antibody.
