Equilibrium potentials of the postsynaptic membrane of frog tonic fibers during changes in the extracellular ionic medium

Abstract

The relation of membrane potential (MP) and amplitude of response to ionophoretic application of acetylcholine (ACh) to synaptic zones of isolated tonic (slow) fibers from the iliofibularis muscle of the frogRana temporaria was studied. The MP was shifted by intracellular polarization of the fiber. This relation is nonlinear in the region of hyperpolarization, mainly because of nonlinearity of the input resistance of the fibers. In experiments on 15 fibers the reversal potential of the response to ACh application, which coincided with the equilibrium potential (Ep) of the postsynaptic membrane, was −8 ±0.7 mv. With a decrease in [K+]n by 80%, Ep was shifted toward the potassium equilibrium potential and its value was −21±1.3 mv. The values of Ep for the postsynaptic membrane of fast and slow fibers evidently differ slightly, but the character of the dependence of Ep on [K+]n is approximately the same for both fast and slow fibers.
