Summary
Hydrazinophthalazines, Phentolamine, Phenoxybenzamine, Nylidrine and Isoxsuprine elicit a drinking response and simultaneous antidiuresis in rats. The effect of Hydralazine and Phentolamine on drinking is potentiated by the neuronal sympathetic blocking agents, Bretylium and Guanethidine.If Guanethidine or Bretylium was administered alone in the same dosage, only minor effects on drinking and diuresis were observed. Control animals given 8 ml water by tube excreted 80% of the fluid load within 2 hours, whilst animals pretreated with the combination of Bretylium and Hydralazine showed complete antidiuresis. The plasma volume was increased by 15% in the pretreated group. Since on examination of the intestines it was apparent that the water had been entirely absorbed, it must be assumed that the major part of the fluid had been transferred to the extravascular compartment. Propranolol blocked the effect of both Phentolamine and Hydralazine on drinking.
