Summary
Some biochemical changes in brains from mink with leucodystrophy have been reported. The cerebrum has been analyzed for myelin lipids without separation of white and grey matter, and a heavy decrease of these lipids has been found in the diseased animals. There seems to be a normal content until the time when the clinical symptoms appear i.e. about 50 days after birth. After this time there is a marked increase of myelin lipids in the normal animals; but this increase does not take place in the diseased animals, and short time before death the content of both cerebrosides and sulphatides are about 25% of the normal values. The decrease in cholesterol and phospholipids are not so marked as in the glycolipids but this must be judged in connection with the fact that the grey substance, which is not involved in the destruction, has a considerable content of these lipids.
Incorporation of S35-labelled sulphate into brain sulphatides, in the diseased animals gave a specific activity about four times higher than normal and this high specific activity was kept at the same level during the 19 days after injection. These results are discussed and taken as expression for a normal synthesis of myelin in the diseased animals, but with destruction of previously formed myelin.
Zusammenfassung
Einige biochemische Veränderungen in Gehirnen von Nerzen mit Leukodystrophie werden beschrieben. Das Cerebrum wurde in bezug auf Myelinlipoide ohne Trennung der weißen und grauen Substanz analysiert; eine starke Lipoidabnahme wurde in den kranken Tieren festgestellt. Offenbar ist ein normaler Gehalt vorhanden, bis die klinischen Symptome erscheinen, das heißt, etwa 50 Tage nach der Geburt. Nach dieser Zeit zeigen die normalen Tiere eine erhebliche Vermehrung der Myelinlipoide; diese Vermehrung findet sich nicht bei den kranken Tieren. Kurze Zeit vor dem Tod ist der Gehalt sowohl an Cerebrosiden als auch an Sulfatiden auf etwa 25% der normalen Werte abgesunken. Die Abnahme des Cholesterins und der Phospholipoide ist nicht so ausgeprägt wie die der Glykolipoide. Das ist vielleicht darauf zurückzuführen, daß die graue Substanz, die nicht zerstört wird, bedeutende Mengen von Cholesterin und Phosphatiden enthält.
I.p. Injektionen von S35-markierten Sulfat ergibt in den Gehirnsulfatiden der kranken Tiere eine spezifische Aktivität, die etwa viermal größer als normal war. Diese große spezifische Aktivität war 19 Tage nach der Injektion unverändert. Die Resultate werden als Ausdruck normaler Synthese von Myelin bei den kranken Tieren interpretiert, die gleichzeitig mit einem Abbau früher gebildeten Myelins vonstatten geht.
