Summary
Female albino rats were used in an investigation of the chronic effects of exercise, consisting of a strenuous program of swimming or treadmill running. The heart ventricles of the exercised animals hypertrophied significantly. Concomitantly, the level of activity of glycerol-P dehydrogenase (EC 1.1.2.1) doubled in heart mitochondria of the exercised rats, and the activity of succinate dehydrogenase (EC 1.3.99.1) increased approximately 40%. The rise of both enzymes' activities apparently was due to a net increase in enzyme protein. This is suggested by the finding of a general increase in mitochondrial protein.
Almost the same results were found for liver mitochondria. Strenuous exercise increased significantly the concentrations of cytochromes and the activities of both glycerol-P dehydrogenase and succinate dehydrogenase.
In a previous paper we had shown that, in a qualitative way, the changes demonstrated here in the heart and liver were evidenced similarly in the skeletal muscle. Since corresponding enzymatic changes (glycerol-P dehydrogenase, succinate dehydrogenase, cytochromes) can be seen in mitochondria after thyroxine treatment, we are inclined to hypothesize that there might be a causal relationship between the effects of physical exercise and the function of the thyroid gland. We will study this presumed relationship next.
Zusammenfassung
Durch intensives Schwimm- und Lauftraining nimmt das Herzgewicht von Ratten signifikant zu. Parallel dazu steigen die Aktivitätsgehalte von Glycerin-1-phosphat-Oxydase (EC 1.1.2.1) und Succinatdehydrogenase (EC 1.3.99.1) um 100% bzw. 40% an. Der Gehalt an Mitochondrienprotein ist im Herzmuskel der trainierten Tiere signifikant größer als bei den untrainierten Kontrolltieren. Die trainingsbedingte Zunahme der Mitochondrienfraktion weist darauf hin, daß der Aktivitätsanstieg der GP-OX und SDH offenbar auf eine vermehrte Synthese von Enzymprotein zurückzuführen ist.
Auch in Lebermitochondrien nehmen durch intensives körperliches Training die Aktivitätsgehalte von GP-OX und SDH signifikant zu. Diese Adaptation des Leberstoffwechsels entspricht in qualitativer Hinsicht den Wirkungen des Trainings auf die energieliefernden Prozesse im Herz- und — wie früher nachgewiesen — im Skeletmuskel. Körperliches Training bewirkt in den untersuchten Organen charakteristische Änderungen der mitochondrialen Enzymausstattung, die in äquivalenter Weise auch nach Applikation von Thyreoidhormonen beobachtet werden. Befunde, die darauf hinweisen, daß zwischen den Wirkungen des Training und der Aktivität der Schilddrüse ein kausaler Zusammenhang besteht, werden in einer folgenden Arbeit mitgeteilt.
