Summary
The pancreases of 25 albino rats, which were fed with condensed milk (=Cudeficient diet) after the suckling-period, were studied. Clinical signs of copper-deficiency observed were: retarded increase in weight, defective hair-covering, discolored incisivors, diarrhea, slight exophthalmos. After 7 or more weeks of Cu-deficient nutrition the pancreas macroscopically shows severe atrophy and lipomatosis. Histologically, the acinar cells have a decreased basophilia, their RNA-content is also decreased, the zymogen-granules are reduced in number, and the nuclei are pyknotic or show karyolysis. Besides these atrophic changes, hypertrophic-degenerative chances develop with giant cells and giant nuclei. The normal acinar structure becomes more and more altered due to a destruction of the cells. Without preceding exudation or cellular infiltration the destroyed cells are replaced by connective-tissue and fat-cells. The findings are thought to be due to disturbances in enzyme-synthesis owing to Cudeficiency, and are classed with the “metabolic lesions of the pancreas” (Doerr, 1964).
Zusammenfassung
Untersucht wurden die Bauchspeicheldrüsen von 25 Albinoratten, die nach der Säugeperiode ausschließlich mit Milch (=Cu-Mangelkost) gefüttert wurden. Der Beschreibung der Pankreasläsionen werden einige klinische Symptome der Cu-Mangelkrankheit vorangestellt: verzögerte Gewichtszunahme der Tiere, Felldefekte, Farbänderung der Schneidezähne, Diarrhoe, leichter Exophthalmus. Die Bauchspeicheldrüsen weisen nach 7- und mehrwöchiger Cu-Mangeldiät makroskopisch hochgradige Atrophie und Lipomatose auf. Histologisch finden sich an den Acinuszellen verminderte Basophilie, RNS-Schwund, Abnahme des Cymogengranulagehalts sowie pyknotische und karyolytische Zellkerne. Daneben treten hypertrophisch-degenerative Veränderungen in Form von Riesenkernen und Riesenzellen auf. Durch Zelluntergänge wird die normale Acinusstruktur in zunehmendem Maße zerstört. Die Defekte werden durch Binde- und Fettgewebe ersetzt, ohne daß es vorher zur Exsudatbildung oder zu cellulärer Infiltration kommt. Die Befunde werden auf Fermentsynthesestörungen infolge Cu-Mangels zurückgeführt und in den Formenkreis der „metabolischen Pankreasläsionen“ (Doerr, 1964) eingereiht.
