Summary
The quaternary methyl and ethyl derivatives of phenylethylamine, and the corresponding ortho-bromo compounds were tested for their biochemical effects (acetylcholine esterase and monoamine oxidase), as well as for their pharmacological actions.
1. True choline esterase (e-esterase) was inhibited by all compounds investigated. o-Bromo-substitution and stepwise replacement of methyl-groups at the quaternary nitrogen by ethyl-groups enhanced the inhibitory potency.
Conversely, inhibitory effects on pseudo-esterase gradually declined with inereasing the number of ethyl-groups, resulting in activation of the enzyme by phenylethyl-triethylamine. Ortho-substitution with bromine only increased inhibitory effects and converted phenylethyl-triethylammonium from an activator to an inhibitor.
2. Monoamine oxidase(MAO). Phenylethylamine, a poorer substrate of MAO than tyramine, became by o-bromination a better substrate than the latter. Phenylethyl-dimethylamine by o-bromination was converted from an inhibitor to a substrate of the enzyme, whereas the inhibitory action of phenylethyl-diethylamine was enhanced. — The quaternary derivatives of phenylethylamine were weak inhibitors of MAO. Regardless of the nature of the N-alkyl-substituents, o-bromo-substitution increased their inhibitory potency.
The effect of halide substitution in ortho-position is readily explained by the formation of eutopic and dystopic enzyme-substrate complexes resp., as proposed by Zeller for iodinated derivatives of benzylamine.
3. Pharmacological effects. In cats phenylethyl-trimethyl-ammonium produced a rise in blood pressure and a contraction of the nictitating membrane caused by stimulation of sympathetic ganglia. In contrast, phenylethyl-triethylammonium had ganglionic blocking properties.
o-Bromination of the trimethyl-derivative reduced its nicotinic (frog rectus) and enhanced its muscarinic actions (blood pressure lowering; stimulation of rabbit ileum). There was also an increase in affinity for “muscarinic receptors” of sympathetic ganglia: not only chlorisondamine but also atropine caused a competitive inhibition of the pressor action.
The ganglionic as well as the neuromuscular blocking effect of the triethyl-derivative was increased by o-bromination.
Zusammenfassung
Die quartären Trimethyl- und Triäthyl-Verbindungen des Phenyläthylamins sowie ihre in ortho-Stellung bromierten Derivate wurden auf biochemische (Beeinflussung der Acetylcholinesterase- und Monoaminoxydase-Aktivität) und pharmakologische Wirkungen untersucht.
1. Die e-Esterase wurde durch alle Verbindungen gehemmt. o-Bromierung und zunehmender Ersatz von CH3- durch C2H5-Gruppen am quartären Stickstoff verstärkte die inhibitorische Wirkung. Umgekehrt bedeutete für die Pseudoesterase der schrittweise Ersatz von CH3- durch C2H5-Gruppen Abschwächung der Hemmwirkung und schließlich (Triäthyl-) Umkehr in eine aktivierende Wirkung. o-Bromierung verstärkte nur die inhibitorische Wirkung und wandelte Aktivatoren (Triäthyl-) in Inhibitoren um.
2. Monoaminoxydase (MAO). Phenyläthylamin, ein schlechteres Substrat der MAO als Tyramin, wurde durch o-Bromierung zu einem besseren als dieses, und Phenyläthyl-dimethylamin aus einem Hemmstoff der MAO zu einem Substrat des Ferments. Die Hemmwirkung des Phenyläthyl-diäthylamins wurde hingegen durch o-Bromierung verstärkt. — Die quartären Derivate waren schwache Fermentinhibitoren. Unabhängig von der Natur der Alkylsubstituenten am Stickstoff, erfuhr ihre inhibitorische Wirkung durch o-Bromierung eine Potenzierung.
Die Auswirkung der Halogensubstitution in ortho-Stellung ließ sich mit den von Zeller für Benzylaminderivate entwickelten Modellvorstellungen von der Bildung eutoper bzw. dystoper Ferment-Substrat-Komplexe zwanglos erklären.
3. Pharmakologische Wirkungen. Phenyläthyl-trimethyl-ammonium wirkte durch die Erregung sympathischer Ganglien blutdrucksteigernd und führte zur Kontraktion der Nickhaut (Katze), während das Triäthylderivat ganglionär blockierende Eigenschaften besaß.
o-Bromierung, die bei der Trimethylverbindung zu einer Abschwächung der nicotinartigen (Froschrectus) und zu einer relativen Verstärkung der muscarinartigen Wirkungskomponente (Blutdrucksenkung; Darmerregung) führte, verlieh dieser auch eine größere Affinität zu den „Muscarinreceptoren“ der sympathischen Ganglien, so daß nicht nur Chlorisondamin, sondern auch Atropin eine kompetitive Abschwächung der blutdrucksteigernden Wirkung verursachte.
Die ganglionär und neuromuskulär blockierende Wirkung des Triäthylderivates wurde durch o-Bromierung verstärkt.
