Summary
The influence of the cardiac glycosides g-strophanthin, digitoxin and lanatoside was investigated on the following properties of excised frog sartorius muscle fibres: resting potential, action potential, water content and intracellular concentrations of sodium and potassium ions.
The three cardiac glycosides at concentrations between 10−8 and 10−6 g/ml produced, after a delay of an hour or more, a slowly progressing depression of the resting potential. This membrane depolarization was not reversible by repeated washings nor was it dose dependent. The average depolarization produced by four to six hours application of the drug was 6–10 mV. However higher concentrations (10−5 g/ml) produced more profound depolarizations as well as occasional spontaneous twitchings in the muscles.
Action potentials produced by direct stimulation of single fibres were little changed apart from a reduction in amplitude after 4 h treatment with glycosides in the concentration range of 10−6 to 10−8 g/ml.
The water content and the intracellular concentrations of sodium and potassium were not influenced by the glycosides in a concentration of 10−7 g/ml applied for as long as 6 h. However, g-strophanthin at a concentration of 10−6 g/ml for 4 h produced a slight decrease in sodium and potassium concentration gradients across the cell membrane and at 10−5 g/ml there was a highly significant reduction in the gradients.
It is concluded that the cardiac glycosides have an effect on the permeability of the resting muscle cell membrane. The relevance of these findings to the therapeutic action of cardiac glycosides on the heart is briefly considered.
Zusammenfassung
Der Einfluß von g-Strophanthin, Digitoxin und Lanatosid A auf das Ruhe- und Aktionspotential, auf die intracelluläre Kalium- und Natriumkonzentration und auf den Wassergehalt isolierter Froschskeletmuskeln wurde untersucht. Es ergab sich, daß die drei Cardenolide im Konzentrationsbereich von 10−8 bis 10−6 g/ml eine verzögert einsetzende und langsam fortschreitende, in diesem Bereich offensichtlich nicht dosisabhängige, irreversible Verminderung des Membranruhepotentials verursachen. Die Potentialsenkung belief sich nach einer Einwirkungszeit von 4 und 6 Std auf durchschnittlich 6 bzw. 10 mV. Höhere Konzentrationen (10−5 g/ml) bewirkten eine verstärkte Membrandepolarisation, die von Spontankontraktionen des Muskels begleitet war. — Das durch direkte elektrische Stimulation von Einzelfasern ausgelöste Aktionspotential der Muskeln zeigte nach vierstündiger Einwirkung der Cardenolide (10−8 bzw. 10−6 g/ml) außer einer der Ruhepotentialsenkung entsprechenden Abnahme der Amplitudenhöhe keine Veränderungen. — Der K+-, Na+- und Wassergehalt der Muskeln wurde durch eine sechsstündige Inkubation mit g-Strophanthin, Digitoxin und Lanatosid A in der Konzentration von 10−7 g/ml nicht beeinflußt. Strophanthin in der Konzentration von 10−6 g/ml bewirkte eine angedeutete, in der Konzentration von 10−5 g/ml eine deutliche Verminderung der K+- und Na+-Konzentrationsgradienten. — Aus den Ergebnissen wird gefolgert, daß die Cardenolide die Ionenpermeabilität der Skeletmuskelzellmembran in begrenztem Ausmaß reduzieren. Dieser Effekt, der den Ablauf des Aktionspotentials offensichtlich nicht beeinträchtigt, ist wahrscheinlich bereits dem subtoxischen Dosenbereich der Cardenolide zugeordnet. Es wird diskutiert, in welcher Weise sich ein derartiger Permeabilitätseffekt der Cardenolide an der insuffizienten Herzmuskelzelle auswirken würde.
