Summary
The production and properties of an antiserum against the RP 2-L-fraction of histones derived from the Walker carcinosarcoma of the rat have been reported. This histone fraction shows a higher turnover in tumour cells than in normal tissues. In the double diffusion test up to 4 precipitation lines could be demonstrated. After injection into rats of Walker tumour cells together with the immune serum, or the injection of cells preincubated with the same serum, no demonstrable differences in tumour growth as compared with the controls have been observed. Also, the addition of the serum to a tissue culture of Walker tumour cells was without effect. First experiments showing that histones of this fraction from rat tissues and from rabbit or calf cells have no antigen in common are discussed in relation to the concept of the Stedmans that histones act as gene inhibitors and regulators.
Zusammenfassung
Es wird über die Herstellung eines Antiserums gegen die RP 2-L-Fraktion von Histonen aus dem Walker-Carcinosarkom der Ratte berichtet. Diese Fraktion zeigt bei Tumoren einen höheren Umsatz als bei Normalgeweben. Im Doppeldiffusionstest wurden bis zu 4 Präcipitationslinien gefunden. Die gleichzeitige Injektion von Walkertumorbrei mit dem Immunserum bzw. die Injektion von mit dem Serum inkubierten Zellen in Ratten zeigte gegenüber den Kontrollen keine Veränderungen der sich entwickelnden Tumoren. Auch der Zusatz des Serums zu einer Gewebekultur von Walkertumorzellen führte gegenüber den Kontrollen zu keinen erkennbaren Unterschieden. Erste Befunde über eine fehlende Antigengemeinschaft von Rattenhistonen und Histonen aus Kaninchen-und Rindergewebe werden im Hinblick auf die Stedmansche Theorie über die Rolle der Histone als Genregulatoren diskutiert.
