Summary
The activities of 13 cytoplasmatic enzymes of 6 different strains of Ehrlich ascites carcinoma have been determined. Some of these enzymes (e.g. glucosamine 6-phosphate deaminase) show only small activity differences from one strain to the next. Other enzymes, exhibit significant differences between the investigated strains. For example, glyceraldehyde phosphate dehydrogenase and phosphofructokinase show activity differences up to 500 per cent.
Activities of α-glycerophosphate dehydrogenase range from 0.25–70.9 μMoles/h/mg protein. an adequate operation of the glycerophosphate cycle does not seem possible in the strain with the low activity. The activities of lactate, α-glycerophosphate, and malate dehydrogenases are interdependent. Strains exhibiting only low activities of glycerophosphate dehydrogenase and malate dehydrogenase are characterized by high lactate dehydrogenase activity and vice versa. The growth rate of the strains parallels their quotient of LDH:GDH.
Zusammenfassung
Die Aktivität von 13 Enzymen der cytoplasmatischen Fraktion von sechs verschiedenen Stämmen des Ehrlich-Ascitescarcinoms wurde bestimmt. Ein Teil der Enzyme ist in allen untersuchten Tumoren in annähernd gleich hoher Aktivität vorhanden. Dies trifft vor allem für die Glucosamin-6-phosphat-desaminase zu. Andere Enzyme hingegen weisen starke Aktivitätsunterschiede auf. Dazu gehören beispielsweise die Clyceraldehydphosphat-dehydrogenase und Phosphofructokinase, deren Aktivität bis zum fünffachen der geringsten gefundenen Aktivität betragen kann.
Die höchste gefundene Aktivität der α-Glycerophosphat-dehydrogenase beträgt 70,9 μMol/h/mg Protein, die geringste 0,25 μMol/h/mg Protein. Beim zweiten Stamm erscheint ein ausreichender Ablauf des Glycerophosphatcyclus nicht gewärleistet. Die Aktivitäten der Lactat-, α-Glycerophosphat- und Malatdehydrogenase weisen nicht nur starke Unterschiede zwischen den verschiedenen Stämmen auf, sondern scheinen auch in gewisser Abhängigkeit voneinander zu stehen: Stämme mit geringer GDH- und MDH-Aktivität weisen hohe LDH-Aktivität auf und umgekehrt. Mit dem LDH:GDH-Quotienten geht auch die Wachstumsgeschwindigkeit der einzelnen Stämme konform.
