Summary
Inosine-Diphosphatase shows a strong cytochemical reaction in the rat adrenal cortex. The distribution of the enzyme is homogeneous throughout the 4 zones of the gland. The intensity and the specificity of the reaction depend on the method of fixation. Short fixation by perfusion gives stronger precipitation with more homogeneous intracellular distribution than obtained by immersion-fixation. The final product of the reaction is localized mostly in the cisternae of the endoplasmic reticulum. The Golgi apparatus is also marked, the nuclear envelope rarely. A positive reaction in the interior of mitochondria seems to be due to incomplete morphological preservation of this organelle.
Biochemically, the activity of the enzyme is important in the homogenate. After differential centrifugation, the greatest part of the total activity was found in the microsomal fraction. In general, there is a good correlation between IDPase activity and the presence of microsomes in the other subcellular fractions. The IDPase seems more firmly attached to the ER-membranes in the rat adrenal cortex than the Glucose-6-Phosphatase, therefore, it should be considered as the best microsomal marker in this organ.

Détectée par une réaction cytochimique, l'enzyme Inosine-Diphosphatase montre au niveau de la corticosurrénale du rat une activité intense, également répartie dans les quatre zones de l'organe. Le précipité spécifique de la réaction se trouve en grande majorité au niveau du réticulum endoplasmique des cellules parencymateuses. Le marquage de l'appareil de Golgi est partiel, celui de l'enveloppe nucléaire rare. La spécificité de la réaction dépend de la préservation des organites. Une fixation uniquement par perfusion perment d'obtenir les résultats les plus favorables quant à l'homogénéité de la réaction. La localisation fine de l'enzyme par rapport aux membranes du réticulum endoplasmique a également pu être étudiée à l'aide de fixation par perfusion. La distribution déterminée par dosage biochimique au niveau des organites subcellulaires, séparés par centrifugation différentielle, montre une très bonne corrélation entre l'activité enzymatique et la présence de microsomes. Il faut tenir compte lors des dosages biochimiques de l'activité phosphatasique acide aspécifique dans chaque fraction, afin d'obtenir cette corrélation.
