Photometrical determination of enzyme activities in living epidermal cells

Summary

A simple apparatus is developed, by which enzymatic measurements in vital epidermis under almost physiological conditions can be carried out. Differentiation between processes with in the epidermis cell (and its wall) and the reaction of the enzyme liberated from the epidermis by diffusion is possible by prosecuting the reaction stepwise.

Kinetics of the lactate dehydrogenase-dependent and the malate dehydrogenase-dependent reaction are presented. The reactions depending on glucose-6-phosphate dehydrogenase could not yet be clearly differentiated.

Zusammenfassung

Es wurde eine einfache Apparatur entwickelt, mit deren Hilfe enzymatische Messungen an vitaler Epidermis in vitro unter weitgehend physiologischen Bedingungen verfolgt werden können. Durch Führen der Reaktion in Stufen kann zwischen Vorgängen in den Zellen der Epidermis (sowie an Zellwänden) und der Reaktion des aus der Zelle diffundierten freien Enzyms differenziert werden.

Dies konnte an den Beispielen der Lactat-Dehydrogenase- und der Malat-Dehydrogenase-abhängigen Reaktion belegt werden. Dagegen waren die Reaktionen in bezug auf die Glucose-6-phosphat-Dehydrogenase bisher nicht eindeutig differenzierbar.
