Isolation of sarcolemma from human skeletal muscle

Summary

An isolation procedure for sarcolemma of human skeletal muscle is described. The method includes the possibility to prepare sarcoplasmic reticulum from the same muscle fibres. Electron microscopy reveals a homogeneous final fraction of 80–90% myofibre enveloping membranes contaminated by blood vessel membranes. The typical three-laminar composition of isolated sarcolemma is demonstrated. The mechanism of muscle fibre emptying is discussed.
