Utilizing fractionated tissue extraction techniques it has been shown that some epidermal enzymes (MDH, NADP-IDH, GOT), in all probability, are characterized by both mitochondrial and extra-mitochondrial localization. On the other hand, other epidermal enzymes seem to be localized exclusively extramitochondrially (GDH) or mitochondrially (GLUDH). These findings are in agreement with the reported localization of these enzymes in other tissues.

The activity of the mitochondrial enzymes of the citric acid cycle in the epidermal homogenate is relatively low (mitochondrial MDH, NAD-IDH, SUDH).

The demonstration of the activity of the mitochondrial NAD-IDH proves that the isocitrate in the epidermal citric acid cycle is oxidized.

By means of quantitative-biochemical methods the following enzymes have been demonstrated in the epidermis: glycerin-1-phosphate-dehydrogenase, glycerin-1-phosphate-oxydase, NAD-isocitrate-dehydrogenase.

In view of a relatively strong glycolysis and a relatively rapid flow of substrates through the pentose phosphate cycle in normal epidermis, the capacity of the citric acid cycle seems to be low.

Durch Anwendung einer fraktionierten Gewebsextraktion konnte für einige Epidermisenzyme (MDH, NADP-spezifische IDH, GOT) eine extramitochondriale und mitochondriale Lokalisation wahrscheinlich gemacht werden. Demgegenüber scheinen andere Epidermisenzyme nur extramitochondrial (GDH) oder nur mitochondrial (GLUDH) lokalisiert zu sein. Diese Befunde stehen mit der von anderen Geweben berichteten Lokalisation dieser Enzyme in Übereinstimmung.

Die Aktivität mitochondrialer Enzyme des Citronensäure-Cyclus ist im Epidermishomogenat relativ niedrig (mitochondriale MDH, NAD-spezifische IDH, SUDH).

Durch den Nachweis der mitochondrialen NAD-spezifischen IDH wird der Schritt der Isocitratoxydation im epidermalen Citronensäure-Cyclus gesichert.

Mit quantitativ-biochemischer Methodik werden folgende Enzyme in der Epidermis (Meerschweinchen) nachgewiesen: Glycerin-1-phosphat-Dehydrogenase, Glycerin-1-phosphat-Oxydase, NAD-spezifische Isocitrat-Dehydrogenase.

Im Verhältnis zu einer starken Glykolyse und einem starken Substratfluß im Pentosephosphat-Cyclus scheint die Kapazität des Citronensäure-Cyclus in normaler Epidermis gering zu sein.
