The sensitivity of the azo- and the H2SO4-reagent for the determination of imipramine and its metabolites on paper chromatograms was increased. The increase in sensitivity with the azo-reagent was about 20× for imipramine, 10× for desmethylimipramine, 10× for didesmethylimipramine and 5× for iminodibenzyl, and with the modified H2SO4-reagent about 20× for imipramine, 10 × for desmethylimipramine, 8× for didesmethylimipramine and 2× for iminodibenzyl.

A method is described to stabilize the colour compound formed by the H2SO4 reagent with the imipramine.

The reaction products of both reagents with imipramine and its metabolites were eluated and investigated spectrophotometrically.

The HCl-eluates are rather unstable with the exception of iminodibenzyl. A characteristic change in colour is observed when eluating iminodibenzyl with methanol.
Since the eluates obtained from the chromatograms with the H2SO4-reaction are unstable they could not be eluated quantitatively. With a new method they are converted into an stable colour compound.

The azo-reagent as well as the H2SO4-reagent are of a high specifity. No reaction was observed with aqueous extracts of various organs of untreated rats.

The direct determination on paper chromatograms with the H2SO4-reagent is the most suitable method if only small amounts of imipramine and its metabolites are available.

Die Empfindlichkeit des Azo- und des H2SO4-Reagenses zum Nach weis des Imipramin und einiger seiner Metaboliten auf Papier-Chromatogrammen wurde gesteigert. Empfindlichkeitssteigerung mit dem Azo Reagens: Imipramin ca. 20mal, Desmethylimipramin ca. 10mal, Didesmethylimipramin ca. 10 mal und Iminodibenzyl. ca. 5 mal. Empfind lichkeitssteigerung mit der modifizierten H2SO4-Reaktion: Imipramin ca. 20 mal, Desmethylimipramin ca. 10 mal, Didesmethylimipramin ca. 8 mal und Iminodibenzyl ca. 2mal.

Es wird ein Verfahren angegeben, den Farbkomplex, den das H2SO4 Reagens mit dem Imipramin bildet, zu stabilisieren.

Die Reaktionsprodukte beider Reagentien mit dem Imipramin und seinen untersuchten Metaboliten wurden aus den Papier-Chromatogrammen eluiert und spektralphotometrisch untersucht.

Die HCl-Eluate des Azo-Reagenses sind mit Ausnahme des Iminodibenzyl recht instabil. Beim Eluieren des Iminodibenzyl mit Methanol kommt es zu einem charakteristischen Farbumschlag.
Die Eluate, die nach der H2SO4-Reaktion aus den Chromatogrammen gewonnen wurden, sind mit Ausnahme des Iminodibenzyl instabil und konnten daher nicht quantitativ ausgewertet werden. Es wird eine Methode angegeben sie in stabile Farbkomplexe zu überführen.

Sowohl das Azo- als auch das H2SO4-Reagens verfügen über eine hohe stoffliche Spezifität. Sie gaben keine Reaktion mit wäßrigen Extrakten aus verschiedenen Organen unbehandelter Ratten.

Zur quantitativen Bestimmung von geringen Mengen Imipramin und seinen Metaboliten in Organextrakten wird der direkte papier-chromatographische Nachweis mit dem H2SO4-Reagens empfohlen.
