Washed mycelium of saprophytically grown Claviceps purpurea decarboxylates L-leucine, but not D-leucine, to iso-amylamine. The reaction was established in an experiment using 14C-labeled leucine. Mycelium incubated for 24 hours with leucine yielded 0.2–0.25 mg of iso-amylamine/g of dry weight of mycelium. These values corresponded to the optimal concentrations of iso-amylamine found in saprophytically grown cultures after 4–5 weeks of growth.

Maximal decarboxylation occurred between pH4 and 7 and at almost saturated substrate concentrations (0.1 mM). There were no detectable differences in decarboxylation rates under aerobic or anaerobic conditions.

Decarboxylation experiments with 15 other amino acids revealed that only D,L-α-aminocaprylic acid, D,L-α-aminoheptanoic acid, D,L-norleucine, and D,L-norvaline were decarboxylated to the corresponding amines.

No decarboxylation could be demonstrated using cell-free extracts.

The biogenetic formation of the ergot amines, iso-amylamine and n-hexylamine by decarboxylation of leucine and α-aminoheptanoic acid, respectively, has been confirmed. The other demonstrated decarboxylations appear to be without biogenetic importance, since neither the amines nor the amino acids were detectable in the ergot cultures. The existence of a neutral amino acid decarboxylase in ergot is assumed.

Separate experiments have shown that ergot cultures are capable of converting acetaldehyde to ethylamine.

Gewaschenes Mycel von Claviceps purpurea decarboxyliert L-Leucin zu i-Amylamin. Die Reaktion konnte in einem Versuch mit 14C-markiertem Leucin abgesichert werden. Bei 24 stündiger Inkubation mit Leucin werden 0,2–0,25 mg i-Amylamin/g Myceltrockengewicht gebildet. Diese Mengen entsprechen den optimalen Amylaminkonzentrationen, die in saprophytisch wachsenden Mutterkornkulturen nach 4-5 Wochen Kulturdauer nachweisbar sind.

Die Decarboxylierungsreaktion verläuft optimal im pH-Bereich von 4-7, bei fast gesättigter Substratkonzentration (0,1 mM Leucinlösung). Zwischen aeroben und anaeroben Inkubationsversuchen ließen sich keine Unterschiede feststellen, so daß ein oxidativer Abbau von i-Amylamin sowie ein mit der Decarboxylierung von Leucin konkurrierender oxidativer Leucinabbau ausgeschlossen werden kann.

Decarboxylierungsversuche mit 15 weiteren Aminosäuren zeigten, daß außer L-Leucin zu i-Amylamin und D,L-α-Aminoheptansäure zu n-Hexylamin, D,L-α-Aminocaprylsäure zu n-Heptylamin, D,L-Norleucin zu n-Amylamin und D,L-Norvalin zu n-Butylamin decarboxyliert werden. D-Leucin wird nicht decarboxyliert.

Decarboxylierungsversuche mit abgetötetem Mycel und zellfreien Extrakten verliefen völlig negativ.

Die biogenetische Entstehung der Mutterkornamine i-Amylamin und n-Hexylamin aus Leucin bzw. α-Aminoheptansäure wird als gesichert angesehen. Die übrigen nachgewiesenen Decarboxylierungsreaktionen dürften biogenetisch ohne Bedeutung sein, da weder die Amine noch die Aminosäuren aus Mutterkornkulturen nachzuweisen waren. Es wird vermutet, daß im Mutterkornmycel eine unspezifische Aminosäuredecarboxylase für neutrale Aminosäuren vorkommt.

Erste Versuche zur Aminbildung ohne Decarboxylierung zeigten, daß Äthylamin im Mutterkornmycel aus Acetaldehyd gebildet werden kann.
