Abstract
The antidotal activities of aquocobalamineacetate and sodium thiosulfate were tested in guinea pigs and cats. The animals were attached to artificial respirators throughout the experiment and were poisoned with a continuous infusion of sodium cyanide solution (4.1 μMol/kg · min NaCN).
The rate of action of each antidote was determined from the time taken for the HCN exhalation to drop below the level of 100 nMol/kg · min in guinea pigs, and to values below 25 nMol/kg · min in cats; the detoxifying capacity of each antidote was determined from the time taken for the HCN exhalation to rise above the said values and the time interval for normal function of heart activity to be restored.
Aquocobalamine was characterized by its rapid rate of reaction in both the animal species; its detoxifying capacity showed, however, according to our expectations, variations corresponding to the applied doses.
The combination of the antidotes aquocobalamine (100 mg/kg) and thiosulfate (500 mg/kg) proved to possess high rate of reaction and a large detoxifying capacity in guinea pigs. Similar results were obtained in cats with antidote doses of 200 mg/kg aquocobalamine combined with 500 mg/kg thiosulfate.
The slow rate of reaction and large detoxifying capacity of thiosulfate were confirmed in our experiments. Its combination with aquocobalamine showed no undesirable change in its antidotal action providing a time interval of 1 min was maintained between the 2 injections.
Zusammenfassung
Aquocobalaminacetat allein und in Kombination mit Natriumthiosulfat wurde an Meerschweinchen und Katzen, die mit Natriumcyanid infundiert (4,1 μMol/kg · min) und künstlich beatmet wurden, auf seine Antidoteigenschaften untersucht.
Die Ermittlung der Wirkungsgeschwindigkeit der jeweiligen Antidotverabreichung erfolgte aufgrund des unterschiedlich schnellen Abfalls der HCN-Exhalation; unter 100 nMol/kg · min bei Meerschweinchen und unter 25 nMol/kg · min bei Katzen. Als Kriterium für die Entgiftungskapazität wurde der frühere oder spätere Wiederanstieg der HCN-Exhalation und das Zeitintervall der anhaltenden normalen Herztätigkeit der Tiere gewertet.
Aquocobalaminacetat zeichnete sich bei beiden Tierspeoies durch hohe Wirkungsgeschwindigkeit aus. Die Größe der Entgiftungskapazität war erwartungsgemäß unterschiedlich, entsprechend der Höhe der Dosierung.
Eine Dosierung von 100 mg/kg Aquocobalaminacetat zusammen mit 500 mg/kg Thiosulfat war beim Meerschweinchen eine Antidotkombination, die eine hohe Wirkungsgeschwindigkeit und eine große Entgiftungskapazität aufwies. Bei Katzen wurden ähnliche Ergebnisse mit 200 mg/kg Aquocobalamin und 500 mg/kg Thiosulfat erzielt.
Die Antidoteigenschaften von Natriumthiosulfat allein, gekennzeichnet durch eine hohe Entgiftungskapazität aber nur eine langsame Wirkungsgeschwindigkeit, wurde in unseren Versuchen bestätigt. Es konnte kein negativer Einfluß dieser Substanz bei der kombinierten Therapie mit Aquocobalamin festgestellt werden, wenn die Verbindungen getrennt, im Abstand von 1 min verabreicht wurden.
