Summary
Freeze-etched heart muscle mitochondria — in contrast to cell nuclei — do not expose the outer surface of the inner membrane. Fractured faces parallel to the outer membrane and across the inner membrane are compared with fractures through oil- and lecithin emulsions. The thickness and a decoration effect along the hydrophilic interface of bimolecular leaflets of lecithin correspond with those of the lipid component in membrane layers. Lecithin layers, however, show a higher degree of order than the lipid component of native membranes. It is suggested that a high concentration of enzymes in the outer compartment gives rise to the formation of a semi-solid gel in contrast to the liquid sols of other cell compartments.
Zusammenfassung
An Herzmuskelmitochondrien wird bei Präparation mit der Gefrierätztechnik — im Gegensatz zum Zellkern — die Außenfläche der inneren Membran nicht freigelegt. Es werden zu der äußeren Membran parallel verlaufende Brüche und Querbrüche zu der Innenmembran gezeigt. Die entstehenden Flächen werden mit solchen in Öl- und Lecithinemulsionen verglichen. Übereinstimmung ergibt sich an Querbrüchen in der Dicke bimolekularer Lipoidschichten des Lecithinmodells und der nativen Membranen sowie in einem Dekorationseffekt an deren hydrophilen Außenkanten. Die Lecithinschichten zeigen einen höheren Grad der Ordnung als die Lipoidkomponente der nativen Membranen. Erhebliche Unterschiede zeigen Bruchflächen in den Ebenen der Schichtungen von Lecithin und von Mitochondrienmembranen.
Es wird vermutet, daß eine hohe Enzymkonzentration im äußeren Mitochondrienkompartiment zur Bildung eines Gels führt, während andere Zellkompartimente flüssige Sole enthalten.
