Summary
The investigation deals with the fine structure of the segmentally organized, paired and contractile lateral vessel of Hirudo medicinalis L., whose layered wall shows the following structural characteristics.

The inner layer consists of a continuous cell lining with areas which flatten down to some 100 Å and are perforated by typical fenestrations. It is a derivative of the coelom, whose individual elements (A-cells) are anchored one to the other by Zonulae adhaerentes and affixed with half-desmosomes to the thin basal lamina. Within a contracted vessel one finds deep endothelial folds between the longitudinal muscles, which reach down to the ring muscles. These endothelial folds might represent reserve structures as well as guiderails for the transportation of material. The form and structure of the A-cells are strongly dependent on fixatives and characterized by strikingly dense vesiculation after OsO4-Bichromat-fixation, while after the application of glutaraldehyde there is extreme flattening of the cells with increased fenestrations and also complicated infoldings of the endothelial cells. Towards the lumen there are on top of the vasothel very electron dense elements (B-cells) which are poor in vesicles and might be interpreted as modified ameboid cells and are meshed with the A-cells by means of slim cytoplasmic processes. Although the two cell types are in close contact to another there are no intercellular attachment devices.

The relatively thick subendothelial connective tissue layer is composed of a network of delicate filaments which have no cross-striation and appear to be imbedded in a finely granulated matrix.

The media is constructed of an inner layer of longitudinal and outer layers of circular muscle cells. The former belong to the so-called lateral type, their nucleus is situated in a hernia-like sarcoplasmic outpocketing, while the latter is considered to belong to the axial type, with its centrally situated nucleus.
Fundamentally both forms possess a fiber-free central sarcoplasmic „medulla“ which is surrounded (crust-like) by thickly packed fibers wound spirally around the longitudinal axis. Each of these fibers is constructed of sarcomers, and due to the composition of the thin and thick filaments one can recognize typical A- and I-bands so that a sliding mechanism seems possible for the contraction.
The sarcotubules dilate under the cell membrane into subsarcolemmal cisternae where the plasmalemma often exhibits complicated foldings („M-complex“) that might serve for some sort of electric stimulation.

The adventitia contains numerous unmyelinated axonbundles which because of their differing sectional aspects can be divided in 3 types. „Type I“ is identified by its complement of mitochondria and microtubuli as well as the presence of membrane-encapsulated masses of corpuscules, that probably contain catecholamine. „Type II“ appears on the other hand electronmicroscopically empty, while „Type III“ is characterized by its unusually size and the obvious myelinlike formations.

We were unable to show typical neuromuscular synapses, for the distance between the effector and the reactive organ (ca. 1,000 Å) is in excess of the usual cleavage. Nevertheless the axons exhibit a series of obvious changes which are due on the one hand to accumulation of thickly packed vesicles in the type I-axons and on the other hand to extensive digitations of the axolemma. Therefore these structural characteristics can be interpreted as substrate of a humoral or electric impulse relay.

Die Wandschichten des segmental gegliederten, paarigen und kontraktilen Seitengefäßes von Hirudo medicinalis L. sind durch folgende Strukturmerkmale charakterisiert:

Die Innenauskleidung besteht aus einer kontinuierlichen, streckenweise bis auf wenige 100 Å abgeflachten und dort von Fenestrationen durchsetzten Zelltapete. Diese ist ein Coelomabkömmling, dessen Einzelelemente (hier A-Zellen genannt) durch Zonulae adhaerentes miteinander und durch Halbdesmosomen mit einer zarten Basalmembran verankert sind. Bei Kontraktion entstehen zwischen den Längsmuskelbündeln tiefe, bis zur äußeren Ringmuskulatur reichende Endothelfalten, die sowohl Reservebildungen als auch Leitschienen für den Stofftransport darstellen dürften. Gestalt und Struktur der A-Zellen sind stark fixationsabhängig und nach OsO4-Bichromat-Fixierung durch eine auffallend dichte Vesikulation gekennzeichnet, während nach Anwendung von Glutaraldehyd extreme Zellabflachungen mit vermehrten Fenestrationen und komplizierte Auffaltungen des Endothels im Vordergrund stehen. Lumenwärts sitzen dem Vasothel sehr elektronendichte, bläschenarme Elemente (= „B-Zellen“) auf, die als modifizierte Amöbocyten gedeutet werden und durch schlanke Fortsätze mit den A-Zellen verzahnt sind. Trotz engsten räumlichen Kontaktes fehlen jedoch interzelluläre Haftstrukturen zwischen diesen beiden Zellformen.

Das relativ breite subendotheliale Bindegewebslager besteht aus einem Filzwerk zarter Filamente, die keine Querstreifung erkennen lassen und in eine feinstgranuläre Matrix eingebettet erscheinen.

Die Media wird aus einer inneren Schicht von längs und einer äußeren Lage zirkulär verlaufender Muskelzellen gebildet. Erstere gehören zum sog. Lateraltyp — ihr Kern liegt in einer hernienartigen Sarkoplasmaausstülpung —, während letztere zum „Axialtyp“ (zentrale Kernlage) gerechnet werden. Grundsätzlich besitzen beide Formen ein fibrillenfreies, zentrales sarkoplasmatisches „Mark“ mit zahlreichen Mitochondrien und spärlichem endoplasmatischem Retikulum, das von dicht gepackten, schraubig um die Längsachse gewundenen Fibrillen rindenartig („kontraktiler Cortex“) umgeben wird. Jede dieser Fibrillen ist aus Sarkomeren aufgebaut, die infolge der Anordnung der in ihnen verlaufenden „dünnen“ und „dicken“ Filamente typische A- und I-Banden erkennen lassen, so daß ein Gleitmechanismus der Kontraktion wahrscheinlich ist. Die Sarkotubuli erweitern sich unter der Zellmembran zu „subsarkolemmalen Zisternen“, in deren Bereich das Plasmalemm häufig komplizierte Auffaltungen („M-Komplexe“) zeigt, die im Dienst elektrischer Erregungsvorgänge stehen dürften.

Die Adventitia enthält zahlreiche Bündel markloser Axone, die in drei Typen eingeteilt werden. „Typ I“ ist durch den Besitz von Mitochondrien und Mikrotubuli sowie das Vorkommen membranbegrenzter, massendichter Korpuskel gekennzeichnet, die wahrscheinlich Katecholamine enthalten. „Typ II“ erscheint dagegen elektronenoptisch weitgehend leer, während „Typ III“ durch seine ungewöhnliche Größe und das Auftreten myelinähnlicher Formationen charakterisiert ist.

Typische neuromuskuläre Synapsenkonnten wir nicht nachweisen; schon der Abstand zwischen Effektor und Erfolgsorgan blieb mit ca. 1000 Å weit oberhalb der üblichen Spaltbreite. Dennoch zeigten die Axone in diesen Bereichen eine Reihe auffälliger Veränderungen. Diese bestanden einerseits in einer Häufung dicht gelagerter Vesikel in den Typ-I-Axonen und andererseits in intensiven Auffaltungen des Axolemms, also Strukturmerkmalen, die als Hinweis auf eine humorale oder elektrische Impulsübertragung gedeutet werden können.
