Summary
Subfornical organ, organum vasculosum laminae terminalis, and area postrema are examined both histologically and histochemically in adult rabbits and rats of both sexes. The results are as follows:

The three organs in question are present in both species. Although the subfornical organ and the vascular organ of the lamina terminalis show in rabbits specific differentiations, the three organs — if compared either with each other or with those of other species — have so many features in common, that from the histogenetic and functional point of view they can be regarded as uniform structures.

As far as the topography is concerned there are no striking differences between the two species. The three organs are in contact with the internal and external spinal fluid system.

The organs are highly vascularized. Connective tissue forms a perivascular space which is thought to be the basis for the lowered threshold of the blood-brain barrier demonstrable in the organs. It is only the vascular organ that shows a regular arrangement of the vessels, which are in functional connexion with the so-called vesicular protrusions. They are regarded as model for the function of the perivascular space. The vacuoles have nothing to do with their contents.

The ependyma that covers the organs is extremely flat. In rabbits silver nitrate reducing granules are regularly found in the ependymal cells of the vascular organ.

Although the impregnation of the fibrous glia reveals astrocytes and “tanycytes”, it is thought that on the whole the circumventricular organs contain glial structures that are not as mature as those of the rest of the brain. Elongated cells, a special variety of the protoplasmic astrocytes (Watermann, 1955), are only found in the subfornical organ of the rat. The chromhematoxylin-positive granules of the subfornical organ and the area postrema belong to microglial cells.

As cytoplasmic ribonucleotides are demonstrable in parenchymal cells, they are regarded as modifications of nerve cells. Furthermore a prominent Golgi complex and processes are found. The metabolism of these cells is extremely high.

It is established that the optically empty vacuoles, which are found only in the subfornical and vascular organs in rabbits, are not artefacts. By histochemical means no specific content is demonstrable. Certain criteria give rise to the suggestion that they originate in parenchymal cells and migrate to the surface where they release their contents into the ventricle.

True neurons are sparse, but a great number of myelinated fibres is present. Special endings are not seen. It is discussed that these fibres may link the circumventricular organs to neural control centres.

The chromhematoxylin-positive granules mentioned before are found only in the subfornical organ, and in exceptionally large quantities in the marginal region of the area postrema. They are demonstrable with all staining methods used for the detection of neurosecretion; nevertheless they cannot be identified with neurosecretion. With histochemical methods it is possible to determine a lipid and a carbohydrate component, but no protein. The granules are regarded as glial elements. The results suggest that in both species the three organs in question have metabolic as well as receptive functions.

Subfornicalorgan, Organum vasculosum laminae terminalis und Area postrema von erwachsenen Kaninchen und Ratten beiderlei Geschlechts wurden histologisch und histochemisch untersucht. Die Untersuchungen ergaben:

Die drei Organe sind bei beiden Tieren vorhanden. Obwohl Subfornicalorgan und Gefäßorgan der Lamina terminalis des Kaninchens besondere Differenzierungen zeigen, haben die drei Organe untereinander bei derselben und im Vergleich mit der anderen Species soviel Merkmale gemeinsam, daß sie histogenetisch und, wie diskutiert wird, auch funktionell als einheitlich betrachtet werden können.

Bezüglich der Topographie bestehen keine wesentlichen Unterschiede bei beiden Tieren. Alle drei Organe haben mit dem inneren und äußeren Liquorsystem Kontakt.

Die Organe sind sehr gut vascularisiert. Die Kapillaren haben einen perivasculären bindegewebigen Raum, der als Voraussetzung der in den Organen nachweisbaren erniedrigten Schwelle der Blut-Hirn-Schranke angesehen wird. Nur im Gefäßorgan ist eine gesetzmäßige Anordnung der Gefäße vorhanden, mit denen die sog. blasigen Vorwölbungen in funktioneller Beziehung stehen. Diese werden als Modell für die Funktion des perivasculären Raums betrachtet. Mit ihrem Inhalt haben die Vakuolen nichts zu tun.

Das Ependym über den Organen ist extrem abgeflacht. In den Ependymzellen des Gefäßorgans des Kaninchens werden regelmäßig Silbernitrat reduzierende Granula gefunden.

Obwohl bei der Imprägnation der Faserglia auch Astrocyten und Tanycyten gefunden wurden, scheint es sich in den circumventriculären Organen im allgemeinen um unreifere Gliaformen als im übrigen Gehirn zu handeln. Langzellen, eine besondere Form der protoplasmatischen Glia (Wätermann 1955) wurden nur im Subfornicalorgan der Ratte gesehen. Zu den Mikrogliazellen gehören die chromhämatoxylinpositiven Granula des Subfornicalorgans und der Area postrema.

Da in den Parenchymzellen cytoplasmatische Ribonucleotide nachgewiesen wurden, werden sie als eine Modifikation von Nervenzellen betrachtet. Ferner wurden Fortsätze und ein kräftig reagierender Golgiapparat gefunden. Ihr Stoffwechsel ist außerordentlich hoch.

Es wurde ausgeschlossen, daß die optisch leeren Vakuolen, die nur im Subfornical- und Gefäßorgan des Kaninchens vorkommen, Artefakte sind. Histochemisch konnte kein besonderer Inhalt gefunden werden. Es ergaben sich mehrere Anhaltspunkte, daß sie aus Parenchymzellen hervorgehen und nach Wanderung an die Oberfläche ihren Inhalt in den Ventrikel abgeben.

Echte Nervenzellen werden nur vereinzelt gefunden, doch sind sehr viele markhaltige Nervenfasern vorhanden. Besondere Endigungen wurden nicht gesehen. Daß durch diese Fasern die circumventriculären Organe mit zentralen Regulationsstellen verknüpft sind, wird diskutiert.

Die erwähnten chromhämatoxylin-positiven Granula kommen nur im Subfornicalorgan und in ungewöhnlich großer Menge am Rande der Area postrema vor. Obwohl sie mit allen „Neurosekretreaktionen“ färbbar sind, können sie mit diesem nicht identifiziert werden. Histochemisch konnte eine Lipid- und Kohlenhydratkomponente, aber kein Eiweißanteil bestimmt werden. Sie werden als Gliagranula betrachtet.
Die Untersuchungsergebnisse lassen für die drei Organe beider Tiere sowohl den Schluß auf eine Funktion im Stoffaustausch als auch auf eine receptive Funktion zu.
