Summary
A structural gene for alanyl-tRNA synthetase has been mapped on the chromosome of E. coli by means fo three temperature conditional mutants. The sites for all three mutations are located between cysC and pheA, the gene order relative to the markers tested being cysC-alaS-nalB-pheA.
A number of mutations suppressing the temperature-sensitive phenotype of one of the alanyl-tRNA synthetase strains has been characterized genetically. In terms of their genetic location three different classes can be distinguished: The first class consists of suppressor mutations mapping between strA and aroE, very close to the spectinomycin locus. The second class comprises mutations linked to the leu region of the chromosome; those revertants which were analysed to possess altered S20 proteins of the 30s ribosomal subunit belong to this group. A third group of suppressors was approximately located between aroE and argG; they are not co-transducible with the strA marker.
